我所张连峰课教授题组,首次利用CRISPR/Cas9技术,建立大鼠条件基因敲除动物模型 (Ma,et al.,Generating rats with conditional alleles using CRISPR/Cas9, Cell Research (2014) 24:122-125.)。规律成簇间隔短回文重复(Clustered regularly interspaced short palindromic repeats,CRISPRs),这是一类广泛分布于细菌和古菌基因组中的重复结构,其能够与一系列相关蛋白(Cas)、前导序列一起,能为原核生物提供对抗噬菌体等外源基因的获得性免疫能力。经改造后的CRISPR/Cas9系统可以作为一种基因工程工具,用于大小鼠等高等哺乳动物的基因组编辑。
